What Passes Through a 12000–14000 Da Membrane
Molecules far smaller than the cut-off, such as water, glucose and iodine, pass through the pores in the wall, while large molecules such as starch stay inside. In the usual practical, a length is filled with a starch and glucose mixture, closed at both ends and stood in a beaker of water. After a while the water outside gives a positive Benedict’s test for glucose but no blue-black colour with iodine, showing that only the glucose has crossed. Because water also moves through the wall, from the more dilute side towards the more concentrated one, the same tubing is used to demonstrate osmosis.
Preparing and Sealing a Length
Cut the piece needed for one bag and soak it in water until it turns soft, then rub one end between finger and thumb to open the tube. Close the lower end with a knot or a clip, fill the bag, close the top and rinse the outside before placing it in the beaker, so that nothing left on the surface is mistaken for material that has crossed. Keep the unused tubing dry and sealed.
