How the System Separates a Sample
A pump drives the liquid mobile phase at high pressure through a column packed with fine particles. The sample is injected into that flow, and each compound in it is held back by the packing to a different degree, so the compounds leave the column at different times and pass the detector one after another as a series of peaks.
Isocratic, Binary and Quaternary Versions
The isocratic system pumps a mobile phase of fixed composition, with samples loaded through a manual injector. The binary system mixes two solvents and the quaternary system up to four, so the composition can change during a run as a gradient; both take samples from an autosampler. The 100 Hz sampling rate records fast, narrow peaks in rapid runs, and 2.5 AU is the absorbance up to which the detector response stays linear.
